The official X (formerly Twitter) account of the manga announced that the 194th and final chapter of the series has been published on the 39th issue of Kodansha's Weekly Shōnen Magazine. The manga began publishing in 2021, which means it was serialised for four and a half years. The 22nd and final volume of the manga will be published on 22 October, 2025.

The author Marcey Naito thanked the fans for their continued support for the last few years. The author of The Quintessential Quintuplets and Go! Go! Loser Ranger!, Negi Haruba also drew a celebratory illustration to commemorate the ending of Tying the Knot with an Amagami Sister. Haruko Iizuka, the character designer for the anime, did the same.

The manga was adapted into an anime which aired from October 2024 to March 2025. Crunchyroll streams the anime worldwide and describes the story as:

Uryu Kamihate is a high school student hoping to enroll in Kyoto University’s medical school. After being taken in by the chief priest at Amagami Shrine, he is told he must marry one of three maiden sisters—Yae, Yuna, or Asahi—to live at, and one day inherit, the Amagami Shrine. Kamihate will need to sweep one of the maidens off her feet in this story of romance, friendship, and fun.
